Sarchu is around 13,500 feet - i don't think it's near to 16,000 ft... with proper hydration and diamox, AMS can be taken care of.

Of course hydration and diamox will be helpful but give your body to naturally acclimatize.
Again as Hitanshu says "Respect AMS, do not underestimate it."

^^ totally agree...! alternate night halt for the second day would be jispa which is at a lower altitude, but the third day's ride would be pretty hard.
personal opinion - on the ride last year, we heard the same advice - nevertheless, we stayed overnight at sarchu. It was ok

1) have to keep revving higher or change gears a lot of times
2) low handlebars add to your worries... if you have to take on tough terrain for over 10 days
3) the rear tyre cannot get grip on rocks - you have many such places on the way.. may be you should get a different tyre...

When we were there we kept ourselves hydrated but we were a little late in taking the medicine (diamox). ^^...totally agree! nothing worse than getting hit by AMS. and yeah - not trying to argue with you, safety is the most paramount factor in the trip. No heroism allowed
