I would be grateful if any of you were to recommend a suitable route in and around Pune/Mumbai which will simulate the road conditions of the Jammu - Leh - Manali circuit as accurately as possible.

The following route is what I'll suggest.
Pune folks take Tamhini route and team up with Mumbai folks at Mangaon on NH17.
Ride till Khed, have lunch & turn back.
At Mahad turn towards Bhor via Varandha ghat.

Riding on NH17 will help you for the 2 lane roads mountain/curvy road you will get from Jammu till Jawhar tunnel. Though the traffic on Jammu till Jawhar tunnel will be more than NH17.
Varandha ghat will help you in riding on bad sections of ladakh. (Not sure how the surface of Varandha right now is but last time when I went there, it was hard surface with some gravel and bumps).

All this if you haven't ridden off good tarmac roads much.

For group riding and getting familiar with other team members riding style, a couple of short rides together on NH4 would do.

You might not want to club both (getting familiar with people's riding style & getting familiar with challenges of riding on 2-lane and hilly roads) at the same time.

At Zojila & some other mountain passes you might have to take care of slush for that wait till the rains arrive and then find out some place that is not having red-slippery mud (like Rajmachi).

Tail Baila is also a good venue for such practice runs. At last weekend's ride, I overheard people saying that it reminded them of Ladakh. Though I am not sure myself since I have never been to Ladakh.

I won't suggest anyone to visit Rajmachi(at the most go till the base from where the really bad sections starts) or Tail Baila(Amby valley - Tamhini is more than enough instead) for preparing to go to Ladakh.
Not sure whom you overheard and where all in Ladakh they have gone to but places in Ladakh visited by most is way better than Tail Baila, if one exclude Zanskar.

Thanks for your inputs.

We wish the ride to be for just one day i.e. start individually at around 0500h from our respective homes, RV at some place en-route to form the team, ride the route and return to be home by about 2000h.

Which of the routes suggested can conform to this time schedule?

Well, he could not react in time on how to go about on a not so tough corner, braked hard in panic and the fork gave away, throwing him off the bike.

He is a experienced rider being on long tours as well, a bad day I guess.

Learning - your mind state should be apt if you are on high speeds.

I didn't quite understand the route; is the Khed you have mentioned the same as Rajgurunagar on the Pune - Nasik highway or is it a different Khed? The route given by you seems to be a bit ulta-pulta if the Khed as per you is indeed Rajgurunagar.
